Reproductive Failure in the Cat
 

SPONTANEOUS OVULATION IN THE CAT
According to a 2005 study at the Department of Reproductive Sciences,Smithsonian Zoological Research Park," Results demonstrate that: (1) both antide and levonorgestrel are effective for inducing short-term suppression of follicular recruitment and ovulation in the cat; (2) inhibition is reversible; and (3) GnRH antagonists and progestins differentially regulate basal estradiol secretion. This study also confirmed a relatively high incidence of spontaneous ovulation in the cat, a species generally considered to be an induced ovulator."
 
REGARDING SPERM QUALITY & TESTING

The majority of wild felids, as well as some domestic cats, have low sperm concentration in their ejaculates, and a high proportion of abnormal spermatozoa. We have employed several possible semen quality markers to further characterize cat epididymal sperm. ..........The annexin V assay revealed a high percentage of sperm with PS exposure, and the TUNEL assay pointed to high levels (13+/-12%) of sperm with DNA breaks. ......... In parallel to this analysis, cat sperm morphology was evaluated using the Diff-Quik optical stain, which has been used in human reproduction laboratories. Several types of abnormalities could be characterized with this method. Remarkably, head staining abnormalities detected using the Diff-Quik staining method were strongly correlated with, and could accurately predict, sperm DNA defects detected in the same sample using the TUNEL assay. We therefore suggest that sperm morphology analysis using Diff-Quik could be used in field conditions to assess sperm status, due to the simplicity of the procedure and the equipment involved.

VIABILITY OF FROZEN SEMEN IN THE DOMESTIC CAT

We examined motility, plasma membrane integrity, and binding capacity to homologous zona pellucidae (ZP) of frozen/thawed epididymal cat sperm as a model species for endangered felines. Epididymal spermatozoa from 20 domestic cats were frozen with freezing egg-yolk extender containing 3.0% glycerol in 0.25-ml straws. Post-thaw motility and plasma membrane integrity of the frozen/thawed spermatozoa were 31.8 +/- 2.4% and 32.2 +/- 4.2%, respectively. The frozen/thawed spermatozoa were co-cultured with frozen/thawed immature homologous oocytes with intact ZP for 3 h to examine their ability to bind to the ZP. Sixteen of the 20 frozen/thawed sperm samples demonstrated the ability to bind to ZP. These results indicated that the freezing system for epididymal sperm used in the present study gives appropriate information for banking the genetic resources of wild felid species.

Many studies have reported new reproductive techniques for reproduction in endangered non-domestic felids. Artificial insemination is an important tool for developing breeding programs. This manuscript reviews recent progress in feline artificial insemination, with particular emphasis on intrauterine sperm deposition. Intrauterine insemination of felids results in higher fertilization rates than intravaginal insemination, despite insemination with fewer sperm. Although surgical insemination was utilized historically, there are some reports of catheterization of the cervix of the queen. Recently, a new technique and catheter were proposed for transcervical insemination of fresh or frozen-thawed semen.

 
ABILITY  OF FERAL CATS TO REPRODUCE: 
RESULTS: Pregnant cats were observed in all months of the year, but the percentage of cats found to be pregnant was highest in March, April, and May. Cats produced a mean of 1.4 litters/y, with a median of 3 kittens/litter (range, 1 to 6). Overall, 127 of 169 (75%) kittens died or disappeared before 6 months of age. Trauma was the most common cause of death. CONCLUSIONS AND CLINICAL RELEVANCE: Results illustrate the high reproductive capacity of free-roaming domestic cats. Realistic estimates of the reproductive capacity of female cats may be useful in assessing the effectiveness of population control strategies.
 
 
 

 

Development of spermatogenic function in the sex maturation process in male cats.

The spermatogenic function and plasma testosterone (T) level in the sex maturation process were investigated as to 180 mixed breed cats ranging from 4 months to 2 years in age to be castrated. Testis/epididymis weights reached a peak at 10 and 8 to 9 months of age, respectively. In the testis, sperm appeared at 5 months of age. At 7 months of age, sperm were observed in 96.2% of the cats. In the tail of the epididymis, sperm appeared in 46.9% of the cats at 6 months of age and in all cats at 8 or more months of age. Furthermore, the mean plasma T level rapidly increased at 8 months of age, and reached a peak (2.64 +/- 0.68 (SE) ng/ml) at 10 months of age. Three of 180 cats (1.67%) had unilateral cryptorchidism. These results suggest that the spermatogenic function in male cats becomes mature at 8 to 10 months of age.

 
REGARDING ERECTILE DYSFUNCTION IN CATS
 
Feline penile erection induced by topical glans penis application of combination alprostadil and SEPA (Topiglan).

The objective of this study was to evaluate the efficacy of topically applied prostaglandin E1 (PGE(1))+5% SEPA (soft enhancement of percutaneous absorption) on the glans penis in a feline erection model. Erectile response after glans penis administration of PGE(1)+5% SEPA cream (Topiglan, MacroChem Co., Lexington, MA, USA) was compared to the erectile response after intracavernosal administration of the triple-drug combination (1.65 mg papaverine, 25 microg phentolamine, and 0.5 microg PGE(1)). The placebo cream and increasing concentrations (0.25%, 2.5 mg/ml; 0.5%, 5 mg/ml; and 1%, 10 mg/ml) of PGE(1)+5% SEPA were applied in a total volume of 0.1 ml via a plastic needle-less syringe. The control triple-drug combination was administrated intracavernosally via a 30-gauge needle at the completion of each experiment to serve as a control reference. With each application of placebo, PGE(1)+SEPA, and the triple-drug combination, changes in intracavernosal pressure and systemic blood pressure were continuously monitored. Topical application of PGE(1)+SEPA induced increases in intracavernosal pressure in a dose-dependent manner, with minimal effects on systemic blood pressure. The increases obtained with 1% PGE(1) Topiglan cream were similar to the intracavernosal pressure values elicited by the standard intracavernosal triple-drug combination. These data demonstrate that topical glans penis application of PGE(1)+SEPA can induce an erectile response in cats with minimal systemic adverse effects. Oral pharmacological agents are the first-line treatment for male ED. Studies investigating the effectiveness of noninvasive modalities such as topical therapy should continue, because these agents have the potential to avoid the systemic effects commonly seen with oral therapies. Additionally, topical therapy may also benefit patients who are unresponsive to oral agents or have explicit contraindications. Topical PGE(1) application to the glans penis may become an important treatment option in selected patients suffering from erectile dysfunction.

Use of Birth Control in the Feline

Many breeders have found it necessary to put their cats on birth control.  This allows the cats to have a break between litters while avoiding Pyometra which is common in the feline who cycles and is not bred.  Perlutex is one of the most effective therapies used by a large number of breeders in the US.  However, this drug is not approved for use in the US and so must be obtained by sources in other countries.  Breeder networking is the most effective way to obtain this drug which runs about a dollar a pill.  The dosage on the pill is one per week.  DO NOT miss a day, as some queens will cycle by missing a single day.  Others can have a day or so late dose and not have a problem.  It typically takes 2 weeks for queens to cycle after removal from medications.  It is recommended that breeders allow the queen to cycle once after removal from medication and then breed on the next cycle.
